The Rb gene is located on chromosome 13q 14 and encodes a nuclear binding protein (p 105-Rb). It exists in the nucleus in the form of activated dephosphorylation and inactivated phosphorylation. Activated Rb protein can inhibit cells from entering S phase from G0/G 1 phase. When the cell is stimulated and starts to divide, Rb protein is phosphorylated and inactivated, which makes the cell enter S phase. When the cell divides into two daughter cells, the inactivated (phosphorylated) Rb protein is regenerated by dephosphorylation, so that the daughter cells are in G 1 phase or G0 static state. If the Rb gene is inactivated due to point mutation or deletion of 13q 14, the expression of Rb protein will be abnormal, and the cell may continue to be in the proliferation period, resulting in malignant transformation. P53 gene is located on chromosome 17. Normal p53 protein (wild type) exists in the nucleus and is activated during dephosphorylation, which prevents cells from entering the cell cycle. Point mutation or deletion of p53 gene was found in some colon cancer, lung cancer, breast cancer and pancreatic cancer, which caused abnormal expression of p53 protein and lost its growth inhibition function, thus leading to cell proliferation and malignant transformation. Recently, it has also been found that some DNA viruses, such as human papillomavirus and SV-40, have carcinogenic effects by binding their cancer proteins with activated Rb proteins or p53 proteins and neutralizing their growth inhibition functions.
