Basic situation of PCR post certificate training
1. Registration time: subject to the notice of each provincial clinical laboratory center.
2. Registration conditions: staff engaged in PCR inspection in PCR laboratory; Other personnel designated by the provincial inspection center.
3. Education requirements: None.
4. Training fee: about 1000-2000 yuan (mainly including theoretical training fee, experimental data fee and data fee); Take care of yourself.
5. Training content: It mainly includes the study of management methods and work guidelines of PCR laboratory, the basic principles of PCR and the progress of related technologies, the quality assurance in the process of PCR, the processing, extraction and preservation of PCR samples, the analysis report of PCR test results and the analysis of common problems, the biological safety and pollution prevention measures of PCR laboratory, and the common problems in the process of acceptance and audit of PCR laboratory.
The specific process of registration and examination of PCR employment certificate
1. The designated institution of the health administrative department organizes PCR training.
Health administrative departments at or above the provincial level (provincial health and wellness committee) designated institutions (provincial clinical laboratory center) organize clinical gene amplification test technology related training.
2. The provincial clinical inspection centers issued a notice to set up PCR training courses.
The provincial clinical inspection centers have issued the Notice on the Training Course for Clinical Gene Amplification Testing Technicians (namely, PCR Employment Certificate) (as shown in the figure). In previous years, the provincial inspection centers were generally held once or twice a year. This year, all the provinces affected by the epidemic have held many training courses related to PCR employment certificates. Taking Beijing as an example, seven PCR training courses have been held this year.
Take part in the PCR certificate training class of the provincial clinical inspection center and pass the exam. The cost varies from province to province, about 800- 1200 yuan, and the provincial temporary inspection centers are held once or twice a year.
PCR: Polymerase chain reaction (PCR) is a molecular biology technique used to amplify specific DNA fragments, which can be regarded as a special DNA replication in vitro. The biggest feature of PCR is that it can greatly increase the amount of DNA. Therefore, as long as a little DNA can be isolated, it can be amplified and compared by PCR, whether it is fossils, the remains of historical figures, or the hair, skin or blood left by the murderer in a murder case decades ago. This is also the power of "trace evidence". This idea was first put forward by Mullis in the United States in 1983, and the simple DNA amplification method was invented by Mullis in 1985, which means the real birth of PCR technology. Up to now, 20 13, PCR has developed to the third generation technology. From 65438 to 0973, Taiwan Province scientist Qian Jiayun discovered the stable Taq DNA polymerase, which also made a fundamental contribution to the development of PCR technology.
PCR (polymerase chain reaction) uses DNA to denature into single strand at 95℃ in vitro. At low temperature (often about 60℃), primers and single strand are combined according to the principle of base complementary pairing, and then the temperature is adjusted to the optimal reaction temperature of DNA polymerase (about 72℃), and DNA polymerase synthesizes complementary strand along the direction from phosphoric acid to pentose (5'-3'). The PCR instrument based on polymerase is actually a temperature control device, which can well control denaturation temperature, renaturation temperature and extension temperature.
