(1) hypertonic culture. Long-term use of antibiotics, lysozyme or the action of complement in vivo often causes the cell wall of bacteria to be destroyed, deleted and mutated into L-shaped bacteria, which needs to be cultured in hypertonic medium. Bacteria can only reproduce when the culture medium contains 15% sucrose or 5% sodium chloride.
It is found in patients with pyelonephritis and bacteremia in the recovery period or recurrence period.
(2) Mycobacterium tuberculosis. The culture method is to leave urine in the morning, separate the heart at 3000 rpm, add 4% sulfuric acid or 2% sodium hydroxide to the precipitate, remove the miscellaneous bacteria after 20 minutes, neutralize and inoculate in Roche culture medium, and report the results after 4-8 weeks.
(3) Fungal culture. If patients use broad-spectrum antibiotics or hormones for a long time, they will often have secondary fungal infections (such as Candida albicans or Cryptococcus neoformans). The urine of these patients was inoculated in Shapiro medium and cultured at 37℃ or room temperature for 5 days for observation.
