Through animal experiments, the effects of Yishen decoction combined with benazepril on improving renal function and reducing TGF-β 1 expression in renal tissue were observed, and the possible mechanism of Yishen decoction on improving renal function was discussed.

Method:

1. 50 SPF healthy adult male SD rats were selected as the research object. After a week of adaptive feeding, 10 rats were randomly selected as the sham operation group (group A), and the remaining 40 rats were only modeled by 5/6 nephrectomy.

2. After successful modeling, 32 rats that met the selection criteria of the study group were divided into group B: model group, 8 rats; Group C: Benazepril group 8; Group d: Chinese medicine group 8; Group E: Yishen combined decoction Benazepril group is referred to as Chinese and western medicine group 8, together with group A, 5 groups.

3. After successful modeling, drug administration was started, and each group was given corresponding drug decoction.

4. Urine protein and blood BUN, Scr, RBC, Hb were detected 24 hours after the experiment, and renal histological observation and TGF-β 1 expression in renal tissue were carried out.

Results:

1. General situation: During the experiment, the rats in the sham operation group showed high alertness, quick response, thick fur, clean luster, no obvious abnormality in growth, consumption and activity, and gained weight; The model group was obviously malnourished, which made it listless, slow in activity, poor in appetite, fluffy in fur and withered in description. Two cases died during the treatment, probably due to renal failure. Compared with sham operation group, rats in benazepril group were depressed, slow in activity and fluffy in fur. The performance of rats with traditional Chinese medicine is similar to that of benazepril group; There is not much difference between WM rats and sham-operated rats in general, but their fur is dry and dark.

2. Effect on blood BUN and Scr: The levels of BUN and Scr in Benazepril group, traditional Chinese medicine group and western medicine group were significantly lower than those in model group (P<0.

But still higher than that of sham operation group; There was significant difference between the integrated traditional Chinese and western medicine group and the traditional Chinese medicine group and benazepril group (P < 0.05). In pathological changes, matrix hyperplasia and glomerulosclerosis can be seen in rats undergoing subtotal nephrectomy, but the degree of glomerulosclerosis in the model group is obviously lighter than that in the treatment group, and glomerulosclerosis is the lightest in the treatment group of integrated traditional Chinese and western medicine; Immunohistochemical results showed that the expression of TGF-β 1 in renal tissue in the treatment group was significantly lower than that in the model group (P<0.

The combination of traditional Chinese and western medicine can reduce TGF-B 1 in renal tissue, which is statistically different from that of traditional Chinese medicine group and benazepril group (P < 0.05). Conclusion: Yishen decoction can reduce the blood BUN and Scr of CRF patients, improve anemia and reduce proteinuria, and can effectively reduce the renal tissue of CRF rats induced by 5/6 nephrectomy through turbidimetry.

Quanduyishen decoction has good anti-renal fibrosis effect, and the curative effect is better after combined with benazepril.
