The publication of these articles has played an important role in promoting the development of life science and precision medicine. Study on the development and functional heterogeneity of human Langerhans cells

In October, 20021/kloc-0, the team of Professor Xu Yao from Dermatology Hospital of China Academy of Medical Sciences, the team of Professor Li Wei from Huashan Hospital affiliated to Fudan University and researcher Li Xiao from Texas Heart Institute in the United States jointly published an article entitled "Arrangement and Interaction of Different Human Langerhans Cell Subsets" in the journal of Immunity. And different developmental regulation is needed.

Based on mass spectrometry flow cytometry (CyTOF) and single cell transcriptome sequencing, the researchers found four subgroups of LC in human epidermal primary LC and umbilical cord blood CD34+ hematopoietic stem cell-derived LC(HSC-LC), and drew a complete trajectory of LC development and differentiation. Through mass flow cytometry (CyTOF), the researchers further studied the changes of LC subsets in psoriasis lesions, and found that the inflammatory response of skin was regulated between LC and T cells through RANKL-RANK and PD-L 1/PD- 1/CD80 axes. This study revealed human LC subpopulations with different phenotypes for the first time, and systematically studied the development and differentiation pathways of different LC subpopulations, as well as the differences of immune responses of different LC subpopulations and the phenotypic and functional changes in psoriasis lesions.

Screening of effective population with persistent asymptomatic infection and latent asymptomatic infection in COVID-19

In this study, the cohort study of asymptomatic novel coronavirus infection period (SSIS) and the integrated mass flow cytometry (CyTOF), transcription sequencing (RNA-seq) and plasma microprotein Olink detection techniques were used to explain the main immunological differences between persistent asymptomatic infection and latent asymptomatic infection from three dimensions: single cell proteome, transcription proteome and plasma proteome. It is found that asymptomatic patients in latent period have unique immunological characteristics, which are mainly manifested as excessive activation and differentiation retardation of monocytes, lymphocyte depletion and immunosuppression. Subsequently, by combining clinical features and transformation, it was found that STC 1 and MMP- 1 levels can better distinguish the two asymptomatic infected people, which is helpful to identify asymptomatic infected people in the early clinical stage. By understanding the mechanism of disease development in COVID-19, this study provides a theoretical basis for early identification of asymptomatic latent infection in clinical work, and early and timely intervention to block its progress and diagnose patients.

Mechanism of microenvironment immunosuppression induced by ILC2 in hepatocellular carcinoma

In this study, based on mass spectrometry flow cytometry (CyTOF), the unconventional ILC2 cell subsets induced by microenvironment of hepatocellular carcinoma were identified, and their relationship with prognosis was studied. Further study found that KLRG 1-ILC2 subgroup showed an increase in the production of chemokines, including CXCL2 and CXCL8, which in turn recruited neutrophils to form an immunosuppressive microenvironment, thus leading to tumor progression and recurrence. This study will help to develop a new immunotherapy for liver cancer with ILC2.

Screening effective population based on disease classification and grading of chondrosarcoma patients

2002165438+February, the team of Professor Ye Zhaoming and Professor Li Binghao from the Department of Orthopaedics, the Second Affiliated Hospital of Zhejiang University published a research paper entitled "Histology of Fresh Tissues Reveals Immunological Classification and Proposes Immunotherapy Candidates for Clinical Tumor Research".

Based on mass spectrometry flow cytometry (CyTOF), this study classified common chondrosarcomas from single cell level for the first time in the world. It is found that there is an "immune failure" subtype in common chondrosarcoma, and all patients who benefit from PD- 1 antibody immunotherapy in clinical treatment meet this subtype. Its mechanism includes that the tumor IDH 1/2 mutation leads to the increase of local chemokine concentration and promotes the homing and recognition of immune cells. Based on clinical problems and clinical needs, this study analyzed the immunological characteristics of rare tumors by observing clinical phenomena and using sufficient clinical resources, and found that some common chondrosarcomas may be sensitive to immunotherapy, and then studied their possible mechanisms, which brought new ideas for systematic treatment of this refractory tumor.

Immunomodulatory effect of bone marrow mesenchymal stem cells on monocytes after acute lung injury

In this study, the dynamic changes of lung immune cells in mice, especially MNPs collected after ALI development and MSC transplantation, were systematically mapped by mass spectrometry flow cytometry (CyTOF) and single cell transcriptome sequencing (scRNA-seq) for the first time. MSC therapy not only regulates the secretion of cytokines and chemokines of MNPs, but also regulates its differentiation and antigen presentation function. This study provides a comprehensive and in-depth understanding of MSC treatment based on MNPs collected from ALI's lung tissue, and provides strong data support for the clinical transformation of MSC treatment of ALI.
