。 Generally, the chromatographic column is filled with vacuum, that is, the stationary liquid is dissolved in acetone, the carrier is added, and the column is packed in vacuum after evaporation. Pay attention to tapping with a rubber hammer when filling. Both ends are stuffed with timely cotton.

。 Aging at higher column temperature before use.

。 If the chromatographic peak widens, the retention time changes or ghost peaks appear, the chromatographic column is invalid.

。 Generally, it is mainly caused by high column temperature and loss of stationary liquid, and it may also be caused by pollution of sample matrix.

Packed columns are not commonly used now. It is recommended to use a thick-bore capillary column with an inner diameter of 0.53mm, which has high column efficiency and good stability.
