What are the applications of enzyme-based biosensors in biomedicine?
The research and application of biosensor is an important way to realize real-time, in-situ and online analysis, and has become one of the research hotspots in analytical chemistry. The immobilization technology of molecular recognition substances has an important influence on biosensors. Immobilization of enzyme is one of the key technologies of enzyme biosensor. In this paper, a new method to improve the activity and stability of immobilized enzyme was studied. In order to obtain a biosensor with excellent performance, the main research work is as follows: Part I: Flow-through chemiluminescence glucose sensor with eggshell membrane as enzyme immobilization carrier. The eggshell membrane is a double-layer membrane inside the eggshell, which is mainly composed of biomolecules and highly crosslinked protein fibers. Has good air permeability and water permeability. In this paper, a novel flow-through chemiluminescence (CL) glucose sensor was developed by using glutaraldehyde as a cross-linking agent and immobilizing glucose oxygen (abbreviation) and horseradish peroxidase (HRP) on eggshell membrane. GOD catalyzes the dissolution of abbreviated sugar to generate hydrogen peroxide, and then in the presence of HRP, the generated hydrogen peroxide oxidizes luminol to produce chemiluminescence. Combined with flow injection analysis technology, glucose was detected by detecting chemiluminescence intensity. The fixing conditions (such as fixing time, GOD/HRP ratio and glutaraldehyde concentration) were discussed in detail. In the past five months, this new biosensor has shown good stability.
