Therefore, it seems really extraordinary to analyze the three-dimensional structure and functional significance of membrane proteins.
There are two important events in the history of membrane protein analysis: one is that Professor Michel analyzed membrane intrinsic proteins for the first time in the world. They gave the three-dimensional structure of the light and reaction center of purple bacteria with a high resolution of 0.32nm, and then explained its function. It can be said that its acquisition is a milestone in the field of membrane protein analysis The other is McKinnon's analysis of ion channel and Agre's analysis of water channel, both of which have made outstanding achievements and won the Nobel Prize.
In 2004, you published an article in Nature about the crystal structure of the main light collecting complex (LHC-II) of spinach:
1) at the resolution of 0.272nm, the crystal structure of the important photosynthetic membrane protein LHC-II was determined, and the third kind of membrane protein crystal was found.
2) A complete LHC-II structural model of protein liposome including membrane protein, pigment molecule and lipid molecule was established, providing nearly 30,000 (29,038) independent and accurate atomic coordinates;
3) Based on accurate structural data, the hot issues of light energy absorption, transmission and light protection in higher plants are discussed for the first time. This is undoubtedly a great contribution to the study of plant photosynthesis. I admire you very much, especially your attitude in doing research. Of course, the analysis of membrane proteins is not easy. At present, the main research is mostly membrane intrinsic proteins, which are characterized by being integrated in the membrane and having amphiphilicity. In order to separate them from the membrane, special detergents and appropriate conditions are needed to avoid inactivation or structural changes. At present, the main methods to analyze membrane proteins are X-ray diffraction, nuclear magnetic resonance and three-dimensional reconstruction of electron microscope. The biggest feature of X-ray diffraction is its extremely high resolution, which can reach 0. 1nm, which is determined by the characteristics of X-ray. Its high resolution determines the importance of X-ray diffraction. Nuclear magnetic resonance (NMR) can be used to show the whole dynamic process of protein conformation change, but it is limited to water-insoluble protein. Of course, the three-position reconstruction technology of low temperature electron microscope invented by Klug has also played a role in some aspects.
In a word, X-ray diffraction is the most important method. Since X-ray was discovered by Roentgen, Prague and his son put forward Prague equation, and then established X-ray crystallography, X-ray has made great contributions to mankind, especially in medical applications. Of course, X-rays also created conditions for dozens of people to win the Nobel Prize.
At present, many people are faced with the problem of how to obtain protein crystals (usually single crystals) which can be used for X-ray diffraction. Single crystal requires high purity, no twin, good orientation and so on.
After analyzing the purple bacterium photosynthetic reaction center, Professor Michel wrote a book about protein's crystallography. Of course, the crystallization process of protein is extremely complicated, which is influenced by many factors, such as protein purity, pH value, temperature, water activity and carrier. Someone has done experiments in low gravity, and the results are very good.
Considering that the crystal structure of protein is mostly maintained by the interaction between water molecules (water content is 30%-70%), and the amphiphilic nature of membrane proteins makes it difficult to form stable and regularly arranged crystals in water, another special solution is proposed to replace water. Crystallography in protein has become a difficult and important subject. I think if anyone can make achievements in this field, its significance is far greater than the analysis of 10 or even 100 membrane protein.
