Application of 1 in drug identification
In HPLC, retention time is related to the structure and properties of components, which is a qualitative parameter and can be used for drug identification. With the development of high performance liquid chromatography (HPLC) and its increasingly important position in Pharmacopoeia, some new methods for drug identification by HPLC have emerged.
For example, the identification of clarithromycin, rifampicin eye drops and seven Chinese medicinal materials of Ilex such as Hong Kong, America and Schisandra chinensis. This undoubtedly provides a new idea for further improving and developing drug identification methods.
2. Application in impurity inspection
Over the years, many pharmacopoeias in various countries have collected the methods of detecting impurities by HPLC, and in the pharmacopoeias promulgated year by year, the types of drugs that can be detected by HPLC have increased. With the continuous development of HPLC technology, many drug analysts at home and abroad have studied and discussed the application of HPLC in the inspection of drug-related substances, and established some new methods.
Such as thienophen hydrochloride, oxazepam and its tablets, quinapril hydrochloride and flutamide. These methods are simple, feasible and concrete.
3. Application in content determination
The content of synthetic drugs and their preparations can be determined by HPLC, which can eliminate the interference of impurities in drugs, additives in preparations and existing drugs. In addition, the HPLC determination methods of cortisone acetate eye drops, quinolones, baicalin in Qinlian oral liquid, dextromethorphan hydrobromide tablets and netilmicin sulfate for injection were also studied in China.
The chemical composition of natural drugs is complex, and there may be one or more effective components, so it is difficult to control the quality of drugs and establish quality standards.
For example, there are many kinds of Siraitia grosvenorii with similar structures. At present, colorimetric method is still the main quantitative analysis method, but the error is large and the result is inaccurate. Chen Weijun et al. established a HPLC method for the analysis of momordica grosvenori saponin, which can be used for the analysis of saponin components in momordica grosvenori saponin extract.
Qi Dan tablets are processed with Salvia miltiorrhiza and Notoginseng Radix as raw materials, which are accepted in the standards issued by the Ministry of Health. The main component of Qi Dan tablets is tanshinone, but there is no method to determine its content. HPLC method was established for the determination of tanshinone in Qi Dan tablets. The ingredients of compound preparation have synergistic effect, and its quality control is difficult, so it is rarely included in Pharmacopoeia.
In recent years, there are many reports on the research of high performance liquid chromatography in this field. For example, the determination of baicalin in compound Houttuynia tablets, the simultaneous determination of metronidazole and fenbufen in Yazhoukang capsules, the rapid determination of sulfacetamide sodium and sulfanilamide in Shaotangling ointment, and the determination of narcotine in compound Ganmao tablets have all achieved ideal results.
