When studying the parts after heat treatment, the samples can be cut at will because of the uniform structure. If you study oxidation, decarburization and surface treatment (such as carburization), you should intercept the sample in the cross section (3): No matter what manual interception method you adopt, you must ensure that the metallographic structure of the observation surface of the sample will not change. Soft materials can be cut by sawing, turning and planing. Hard materials can be cut by water-cooled grinding wheel slicer and EDM. Hard and brittle materials (such as white cast iron) can also be obtained by hammering. For the sample whose surface treatment depth is to be measured, the cutting surface should be perpendicular to the infiltration surface. When studying rolled materials, such as the shape and type of inclusions, the degree of deformation of materials, the degree of grain elongation and banded structure, longitudinal samples should be taken parallel to the rolling direction. In order to study the surface defects of materials and the distribution of nonmetallic inclusions, transverse samples should be cut in the vertical rolling direction. The ideal shape of metallographic samples is cylindrical and square, depending on the specific situation. The general height is 10 ~ 15 mm, and the diameter is φ1o ~15 mm; The side length of a square sample is10 ~15 mm. In practical work, due to the variety of materials and parts, it is difficult to intercept the ideal shape and size of materials and parts, which can generally be determined according to the actual situation. However, it is appropriate to take the height of the sample as half of its diameter or side length, and the shape and size are convenient to hold in your hand for grinding. (4) Sample grinding: the purpose of manual polishing.
Rough grinding: soft materials can be leveled with a file by hand, and general materials can be leveled with a grinder. During operation, the side of the grinding wheel should be used to ensure the level of the sample. It should be noted that the contact pressure should not be too high, and the water should be continuously cooled to prevent the internal structure from changing due to temperature rise. Finally, when chamfering, it is necessary to prevent the sandpaper from being cut during fine grinding. However, it is necessary to observe that the surface samples such as decarburization and carburization cannot be chamfered, and sometimes electroplating is used to prevent the edges of these samples from chamfering. After the rough grinding is completed, all samples should be chamfered.
In the later process, sandpaper or polished objects will crack. It may even be caught by polished objects and thrown out, causing accidents.
Fine grinding: The purpose of manual fine grinding is to eliminate the deep and coarse grinding marks left by rough grinding and prepare for polishing. Fine grinding itself involves multiple operations, that is, from coarse to fine on each number of sandpaper. Hand-finished abrasives are sandpaper, which is divided into various specifications and numbered according to the size of abrasive particles. Manual hand grinding method is to use flat glass plate or flat iron plate to push grinding on metallographic sandpaper. In order to ensure that the test surface of the sample is flat and does not generate arc, the force applied to the grinding surface should be balanced and the grinding surface should be in complete contact with sandpaper. At the same time, the grinding should be carried out in one direction, and the sample should be lifted away from the sandpaper when pushing forward. Fine grinding generally starts from No.0 (w40), and fine sandpaper is changed one by one. Generally, steel samples are polished into No.04 sandpaper, and soft materials such as aluminum and magnesium can be polished into No.05 sandpaper. When replacing fine sandpaper, you should clean the glass plate under the mat to prevent coarse sand particles from falling into fine sandpaper, and at the same time, rotate the grinding direction by 90 to observe whether the last grinding marks are worn off. When finely grinding soft metallographic samples, non-ferrous metals such as aluminum, magnesium and copper should be coated with a layer of lubricant on sandpaper to prevent sand grains from being embedded in soft metallic materials and reduce surface tearing. Commonly used lubricants are engine oil, paraffin, gasoline solution, gasoline and saponified matter.

Chemokines, as a member of cytokine superfamily, are a kind of small molecule protein with directional chemotaxis to cells, which can attract immune cells to participate in immune response and immunopathological process [12]. Up to now, at least 50 chemokines have been found, which are composed of 70- 125 amino acids and have a molecular weight range of 8- 1 1KD. Most chemokines contain two characteristic disulfide bonds formed by four conserved cysteines, so they have very similar high-level structures. C-terminal is α-helix, N-terminal is irregular, and it has high biological activity and binds to receptor.
Chemokines were first discovered as chemokines, but with the in-depth study of chemokines, it is now recognized that chemokines not only have chemotactic effects on white blood cells, but also play a wide range of physiological and pathological roles in the development of immune cells and organs, immune response process, inflammatory reaction, pathogen infection, wound repair, tumor formation and metastasis [14- 16]. With the in-depth study of chemokines and their receptors in recent years, many scholars at home and abroad have found that their combination plays a role in the recruitment and chemotaxis of T cells in autoimmune diseases, and the corresponding T cells gather in target organs to cause tissue and organ damage. Wensel et al. found that CXCL 10 combined with its receptor CXCR3 can directionally recruit cytotoxic T cells into the target organs, and eventually lead to skin damage. Barone et al. showed that the expressions of CXCL9, CXCL 10, CXCL 1 1 and receptor CXCR3 in corneal epithelial cells of lacrimal gland in patients with Sjogren's syndrome (SS) all increased. Goulvestre et al. found that the expressions of CCL2 1, CXCL 12, CXCL 13 and CCL22 increased in thyroid tissues of patients with autoimmune thyroid diseases (AITD), among which CXCL 12 was formed by ectopic secondary lymphoid follicles in thyroid tissues of autoimmune diseases.

Chemokines play a role in recruiting and chemotactic T cells to target organs in autoimmune diseases, but there are few studies on their specific functions in Kawasaki disease. At present, there are few studies on the expression and significance of chemokine CCL5 and its receptor CCR5 in Kawasaki disease, lacking the support of relevant literature and data guarantee. At the same time, the content and extraction method of this study are relatively complicated. Therefore, we need to invest more energy and time in the research to ensure the rationality and accuracy of the research results, and at the same time, we need to further collect and sort out the literature, especially foreign literature, to lay the foundation for this research.
(5) Possibility of completing all papers on schedule.
At present, some research objects have been collected, and the corresponding grouping and blood collection have been carried out. At the same time, domestic scholars have studied the expression of CCL5 and CCR5 in other diseases, which can provide corresponding research methods and route reference. However, the authors have isolated PBMC and successfully activated and proliferated T cells.
